according u.s. census bureau, county has total area of 557 square miles (1,440 km), of 544 square miles (1,410 km) land , 12 square miles (31 km) (2.2%) water.


most of western portion of colquitt county, west of moultrie , state route 33, located in upper ochlockonee river sub-basin of larger ochlockonee river basin, exception of northwestern corner of county, between sale city , west of doerun, located in lower flint river sub-basin of acf river basin (apalachicola-chattahoochee-flint river basin). narrow central portion of colquitt county, running north south of moultrie, , widening occupy gap between u.s. route 319 , state route 133, located in withlacoochee river sub-basin of suwannee river basin. eastern portion of county located in little river sub-basin of same suwannee river basin.
